A large number of studies have focused on plant-derived Extracellular Anti-melanogenic as an alternative to chemotherapeutic agents with non side effects. In the current study, we investigated the anti-melanogenesis effect of extracellular vesicles (EVs) extracted from the leaves and stems of S. japonica. Using spectrophotometric and biochemical methods, we found that leaf-derived extracellular vesicles (LEVs) and stem-derived extracellular vesicles (SEVs) decreased melanin content in a concentration-dependent manner and in the B16BL6 mouse melanoma cell line. Tyrosinase (TYR) activity. It was further confirmed from a large number of analyses and studies that LEV and SEV caused a concentration-dependent reduction of melanin content in melanoma cells. Compared to arbutin, which was used as a positive control, both LEV and SEV had a greater whitening effect on melanoma cells, while LEV produced a greater effect.

Notably, neither LEV nor SEV caused significant cytotoxicity. We also examined the effect of plant-derived extracellular vesicles on the expression of tyrosinase-related proteins (TRPs) in melanoma cells. LEV inhibits the expression of melanogenesis-related genes and proteins, including microphthalmia-related transcription factor (MITF), TYR, TRP-1 and TRP-2. In a human epidermal model, LEV had a stronger inhibitory effect on melanin production than arbutin. Overall, our data suggest that LEV from yellow lacquer wood is a novel natural substance that can be used as an anti-melanogenic agent in medicinal formulations.
